It is made up of four amino acid fragments (Met-Glu-His-Phe) and a Pro-Gly-Pro motif attached to the C-terminus.

The Met-Glu-His-Phe fragment is part of all melanocortin sequences, including adrenocorticotropic (ACTH), but it has no relationship with the hormone’s endocrine effects. 

Whereas the Pro-Gly-Pro motif provides Semax with increased stability so it can pass the blood-brain barrier and reach the central nervous system.

As we talked about earlier, Selank helps with anxiety, depression, and other mood disorders.

In a study by Russian scientists, patients with pre-existing anxiety disorders treated with Selank experienced fewer anxiety symptoms:

“Sixty-two patients with generalized anxiety disorder (GAD) and neurasthenia were studied. The effect of selank (30 patients) was compared to that of medazepam (32 patients).

The patient’s state was assessed with psychometric scales (Hamilton, Zung, CGI). Enkephalin activity in the blood serum was measured as well. 

The anxiolytic effects of both drugs were similar but selank had also antiasthenic and psychostimulant effects. 

The clinical-biological study revealed that patients with GAD and neurasthenia had a decreased level of tau(1/2) leu-enkephalin which was correlated with disease duration, severity of symptoms related to anxiety and asthenia and autonomic disorders. 

The increase of this parameter and stronger positive correlations with anxiety levels were observed during the treatment with selank mostly in patients with GAD.”

In another study involving patients with phobic anxiety and somatoform disorders, those who took Selank experienced reduced anxiety levels lasting for a week alongside an overall improvement in their quality of life.

“A comparative study of the anxiolytic effect and tolerability of selank and phenazepam was carried out in 60 patients with phobic anxiety- and somatoform disorders were examined. 

Pronounced anxiolytic and mild nootropic effects of selank were demonstrated. The anxiolytic effect lasted for a week after last receiving the peptide. 

Selank had a positive impact on the quality of life of the patients.”

In a study published in Neuroscience and Behavioral Physiology, mice given 300 μg/kg of Selank had experienced improvements in memory and learning abilities:

“Selank administration (300 micrograms/kg) significantly activated the learning process in rats with initially poor learning ability compared to piracetam.

The actions of the synthetic heptapeptide preparation Selank on learning and memory processes in rats with initially low levels of learning ability were compared with those in normal rats, using a method based on acquisition of a conditioned active avoidance reflex, with repeated administration of peptide 15 min before the start of training sessions for four days. 

The effects of Selank (300 microg/kg) were compared with the effects of the nootrope piracetam (400 mg/kg). 

These experiments showed that Selank significantly activated the learning process in rats with initially poor learning ability, with effects apparent after the first dose on training day 1. 

The effect progressively increased on repeated administration of Selank: the total number of correct solutions increased and the number of errors decreased. 

The maximum optimizing activity of Selank on learning in normal rats was seen on day 3 of repeated administration and training, i.e., after the formation of the initial consolidation phase.”

According to the Journal of Bulletin of Experimental Biology and Medicine, Selank stabilizes enkephalin levels, which helps reduce stress and anxiety. 

Once again, this improved learning capabilities in mice given the peptide:

“Comparative study of plasma activities of enkephalin-degrading enzymes in mice with different phenotypes of emotional and stress reactions revealed significant differences between intact BALB/c and C57Bl/6 mice by the half-life of plasma leu-enkephalin.

Selank in a dose of 100 micrograms/kg produced an anxiolytic effect in the open-field test and increased the half-life of plasma leu-enkephalin in BALB/c mice, but had no effect on behavioral reactions and enkephalinase activities in C57Bl/6 mice. 

Our results suggest that the anxiolytic activity of Selank is associated with the inhibition of enkephalin-degrading enzymes.”

Apart from its anti-anxiety properties, Selank can also boost the immune system by promoting the production of immune system cells in the body.

Patients suffering from depression who were given Selank had increased IL-6 levels after 14 days of use.

“The experiments in vitro revealed that the drug in concentration 10-7 M completely suppressed gene expression by peripheral blood IL-6 of patients with depression but not of the healthy controls. 

At the same time, a significant increase in IL-6 concentration was observed in the cell culture of peripheral blood of patients in the presence of selank. 

The changes in the Th1/Th2 cytokine balance in vivo were found in the serum of patients with generalized anxiety disorder and neurasthenia who received Selank for 14 days. 

The dynamics of these changes had a significant inverse correlation dependence. 

The cytokine-regulating effects revealed in the study suggest that selank can be used as a novel immunomodulator in patients with anxiety-asthenic disorders. 

Additionally, the adaptogenic properties of selank may be beneficial to its use in elderly people and people exposed to environmental stressors for the prevention of infectious diseases.”

Believe it or not, Selank can help improve symptoms of alcohol withdrawal.

In another study published in the Bulletin of Experimental Biology and Medicine, researchers discovered that Selank can help improve anxiety levels associated with alcohol withdrawal within 48 hours:

“We studied the effects of selank on the development of symptoms of acute 48-h alcohol withdrawal in outbred rats drinking 10 % ethanol as the only source of fluid for 24 weeks. 

In alcohol-preferring animals (mean daily ethanol intake >5.0 g/kg) allowed the free choice between 10 % ethanol and water. 

A single intraperitoneal injection of selank in a dose of 0.3 mg/kg eliminated anxiety induced by ethanol withdrawal in tests elevated plus maze and social interaction tests and prevented the formation of mechanical allodynia without affecting ethanol consumption. 

The findings suggest that selank is effective in eliminating alcohol withdrawal symptoms in rats.”

Moreover, growing research suggests Selank has antioxidant properties supporting liver health.

Rats given 100 and 300 μg/kg of Selank showed improved free radical levels in their liver:

“We studied the effect of Selank administered intraperitoneally in doses of 100, 300, and 1000 μg/kg to male Wistar rats 15 min prior to restraint stress on the content of aminotransferases and total protein concentration in blood serum and intensity of free radical oxidation in the liver. 

Under conditions of acute restraint stress, Selank in doses of 100 and 300 μg/kg decreased catalase and superoxide dismutase activities and malondialdehyde concentration and increased total antioxidant activity in the liver homogenate.

Administration of Selank in a dose of 1000 μg/kg reduced the content of aminotransferases in blood serum, decreased superoxide dismutase activity in the liver, and increased total antioxidant activity. 

Under conditions of chronic stress, Selank in all doses produced similar effects: reduced superoxide dismutase activity and malondialdehyde concentration in the liver tissue and AST activity in the serum. The other parameters remained unchanged.”

But going back to cognitive health, emerging research supports Semax’s beneficial effects on brain function.

In healthy men and women, those given intranasal Semax improved the brain’s default mode network function.

The brain’s default network is a group of regions that are more active when you’re resting compared to when you’re focusing on a specific task

This network is believed to help with internal thoughts, like remembering the past, planning for the future, and daydreaming.

“The effects of the nootropic drug Semax on the neuronal network of the brain were studied by resting-state functional magnetic resonance imaging (resting state fMRI). 

The study was carried out on two groups of healthy volunteers (11 men and 13 women aged 43.9±9.5 years). 

Resting-state fMRI was carried out 3 times: directly before and 5 and 20 min after intranasal 1% Semax (14 subjects) or placebo (10 subjects). 

The topography of the resting state default mode network was studied. 

A greater volume of the default mode network rostral (medial frontal cortex) subcomponent was detected in the Semax group in comparison with controls. 

Resting-state fMRI confirmed Semax’s effects on the neuronal network of the brain and demonstrated the topography of these effects.”

In another study published in the Journal of Aviakosm Ekolog Med, Russia, Semax helped treat memory disorders in white mongrel rats via elevating blood supply to the brain: 

“Experiments with white mongrel rats (both females and males) showed that nootropic substances, i.e. sodium hydroxybutyrate, pyracetam, oxyracetam, aniracetam, centrophinoxine, nooglutyl, mexydol, semax, amide L-pyroglutamyl-D-alanine, and MK-801, a specific noncontesting antagonist of the NMDA-receptor complex, significantly increased survivability of the animals following bilateral occlusion of the common carotid arteries. 

The nootrops prevented, partly or completely, mnestic disorders in the experimental rats. 

In contrast, MK-801 profoundly aggravated these disorders. Similar results were obtained with a model of hypoxic amnesia. 

Except for N-acetylglycine amide and amide L-pyroglutamyl-D-alanine, the nootrops fully or to a considerable extent blocked the development of mnestic disorders in hypoxic rats. 

The authors recommend novel nootrops (nooglutyl, mexidol, semax and GVS-111) for the pharmacological correction of mnestic disorders consequent to hypoxia or cerebral ischemia.”

Semax is known for its neuroprotective properties, which help protect the brain from different types of damage and degeneration. 

One key way it does this is by reducing oxidative stress that can cause damage and death to brain cells. 

By lowering oxidative stress, Semax may slow down the progression of neurodegenerative conditions and protect brain cells from harm:

“Semax is the first domestic nootropic drug of an unexhausted type from the group of neuropeptides. In experimental studies it showed angioprotective, antihypoxic and neurotrophic activity in the doses 100-150 micrograms/kg. 

A combined clinical-electrophysiologic study revealed its high efficiency in acute ischemic stroke.

A clinical trial was performed of immunobiochemical mechanisms of neuroprotective properties of Semax in acute period of ischemic stroke. 

A retrospective comparative clinicoimmunobiochemical analysis provided objective data on the molecular level on activating influence of Semax on antiinflammatory postischemic reactions in the brain. 

Shifting neuromediatory balance toward a prevalence of the antiinflammatory agents (interleukin-10, tumor necrosis factor-alpha) over the factors maintaining the inflammation (interleukin-8, C-reactive protein).”

Semax may also assist in the recovery process following traumatic brain injuries (TBIs), which usually lead to cognitive issues and neurological problems. 

Semax’s ability to protect and restore the brain may aid in the healing and rebuilding of damaged brain tissue.

In fact, in 110 patients who had an ischemic stroke, those given Semax had boosted BDNF levels, improved motor performance, and faster functional recovery:

“One hundred and ten patients after IS (43 men, 67 women, mean age 58.0±9.7, Ме 63 years) were examined. 

All patients were divided into early and late rehabilitation groups. Each group was subdivided into semax+ and semax- subgroups. 

Standard regimen of semax included 2 courses (6000 mcg/day) for 10 days with 20 day interval. 

Plasma BDNF levels, motor performance on the British Medical Research Council scale and Barthel index were assessed in all groups.

Administration of semax, regardless of the timing of rehabilitation, increased BDNF plasma levels which remained high during the whole study period. 

In semax- subgroups high BDNF plasma levels were positively correlated with early rehabilitation. 

Administration of semax and high BDNF levels accelerated the improvement and ameliorated the final outcome of Barthel score index. 

There was a positive correlation between BDNF plasma levels and Barthel score, as well as a correlation between early rehabilitation and motor performance improvement. 

The correlation between BDNF plasma levels and Barthel score was modified by the timing of rehabilitation.”

Apart from its cognitive benefits, Semax has anti-anxiety and antidepressant effects as well. 

Rats that were injected with Semax showed anti-anxiety and antidepressant effects after 14 days of treatment. 

This is due to the activation of the serotoninergic system in the brain and the boost of BDNF expression in the hippocampus:

“Effects of chronic intranasal administration of ACTH(4-10) analog Semax (MEHFPGP) on exploratory activity, anxiety level, and depression-like behaviour were studied in white rats. 

The peptide was injected daily in dose 0.05 mg/kg during 10 or 14 days. 

It was shown that chronic Semax administration at 1-2 weeks induced anxiolytic and antidepressant effects but did not influence the exploratory activity in a non-stressogenic environment. 

The Semax effects may be the results of activation of the brain serotoninergic system as well as increased BDNF expression in the rat hippocampus.”

As part of the ACTH-derived peptide family, Semax can be used to treat ischemic stroke in patients 28 days after a heart attack.

In mice, Semax helps improve heart function and reduces the risk of heart failure after a heart attack.

It also reduces excessive NO production and neurological problems in rat brains caused by reduced blood flow:

“Semax (150 microg/kg) was given i. p. in the operating day twice: 15 min and 2 hours after coronary occlusion, and once a day for the following 6 days. 

In 28 days after infarction, the MI group developed cardiac hypertrophy, cell growth was caused mainly by the increase of contractile filaments not supported by the appropriate mitochondrial growth that indicated an impaired energy supply of the cells. 

Moreover, cardiac hypertrophy was accompanied by decreased mean arterial blood pressure and cardiac contractile function and increased left ventricular end-diastolic pressure. 

Pharmacological change of cardiac afterload revealed that, in 28 days after MI, the rat heart was not able to change its contractile performance in response to either increase or decrease of systemic blood pressure, and as a result could not maintain its diastolic pressure. 

All these changes obviously reflect development of heart failure. 

Semax did not affect cardiac work but partially prevented end-diastolic pressure growth in left ventricle as well as ameliorated cardiomyocyte hypertrophy and disproportionate growth of contractile and mitochondrial apparatus, thus exerting beneficial effect on the left ventricular remodeling and heart failure development late after myocardial infarction.”

Not many people know Semax can increase the quantity and movement of immune cells, as well as upregulate the expression of genes related to immune function.

Semax has applications in treating stroke as it helps protect the brain from damage and improves recovery, thereby reducing the risk of future strokes. 

It works by enhancing brain function, reducing inflammation, and promoting the repair of brain cells.

In fact, in a study with 187 patients who had cerebrovascular insufficiency (a condition where brain arteries are blocked, reducing blood flow), treatment with Semax slowed the disease’s progression and lowered the risk of stroke.

“One hundred and eighty-seven patients with different stages of cerebrovascular insufficiency (CI) have been examined. 

A diagnosis of CI was based on the results of neurological and neuropsychological study, ultrasonic dopplerography, rheo- and encephalography, electrocardiography, brain MRI and eyegrounds examination. 

Neurological scales were used for neurological status assessment and further data processing. 

The study aimed at evaluation of tolerability and clinical efficacy of the medication and complications in CI course. 

Semax treatment resulted in significant clinical improvement, stabilization of the disease progress and reduced a risk of stroke and transitory ischemic attacks in the disease course.

Semax vs Selank Side Effects

Semax and Selank are generally safe when used as directed with few reported side effects. 

Other biohackers who’ve tried Semax have shared some of their experiences with using the peptide.

Some have experienced headaches when exceeding the recommended dosage, although one individual found that their headache went away within 30 minutes after mistakenly taking a 24 mg dose

While there hasn’t been enough human research on Selank to establish an official dosing schedule, a book published by Dr. William Seeds in 2020 has proposed a protocol based on available data and anecdotal evidence from patients using peptide therapy. 

Starting Dose:

  • Begin with the lowest effective dose, typically 300 mcg per day for subcutaneous injection.
  • Take 750-1,000 mcg once a day via nasal spray, spread out over several doses if needed.
  • Monitor your response and adjust the dose gradually by 100 to 250 mcg every few days if needed.

Nasal Administration:

  • Daily doses can go up to 1,000 mcg without any reported side effects.
  • For doses higher than 300 mcg, split the dose into multiple administrations throughout the day.

Dosage Schedule:

  • Administer Selank for 12 weeks, followed by a 4-week break.
  • This cycle can be repeated as necessary.

Take Selank consistently for several weeks or months to assess its effectiveness.

In Russia, two formulations of intranasal Semax have been approved: a 0.1% solution and a 1% solution, each with specific indications and dosage guidelines.

0.1% Semax Nasal Drops:

  • Mental Exhaustion: 400-900 mcg daily, divided into 2-3 doses for 3-5 days.
  • Cognitive Issues in Children and Teenagers (7-18 years): 200-400 mcg daily, split into two doses for 30 days.
  • Optic Nerve Ailments: 600-900 mcg daily, divided into 2-3 doses for 7-10 days.

The 0.1% formulation can be administered daily for up to 30-day cycles, especially in patients with glaucoma.

1% Semax Nasal Drops:

  • Mild-Moderate Stroke: 2,000-3,000 mcg, taken 3-4 times daily at 3-4 hour intervals for 10 days, totaling 6,000-12,000 mcg daily.
  • Severe Stroke: 3,000-4,000 mcg, taken 4-6 times daily at 2.5-3 hour intervals for up to 10 days, totaling 12,000-20,000 mcg daily

For mild-moderate stroke patients, a typical regimen involves a 10-day active period followed by a 20-day washout and then another 10-day active period.

Experimental Research Dosage (0.3% Semax Nasal Spray):

  • Initial Dose: 600 mcg daily (2 pumps).
  • Adjustment: Increase the dose based on individual response, taking up to 900 mcg daily (3 pumps).
  • Study Duration: Up to 30 days for doses higher than 900 mcg daily, or 600 mcg daily for up to 60 days.
  • Cycle: Ensure a rest period equivalent to the study duration following the experiment.

Selank is ineffective when taken orally because it is quickly broken down in the stomach, making the nasal spray an effective method of administration.
