PRACTICALLY speaking, we are far away from the future of effective muscle-building peptides.

Ipamorelin is a growth hormone-releasing peptide (GHRP) that mimics natural growth hormone and ghrelin release. 

As a pentapeptide, it is highly selective and does not impact other hormones like acetylcholine or cortisol.

Compared to other GHRPs, it is a safer and more effective option for enhancing athletic performance while addressing growth hormone deficiencies.

When injected, Ipamorelin stimulates the pituitary gland to release growth hormone, which supports muscle growth while preventing bone/cartilage issues. 

Ipamorelin also boosts cell synthesis, insulin production, and ghrelin production.

All of which promote fat loss and increase lean muscle mass development.

These potent muscle-building properties are the reasons why most bodybuilders opt for Ipamorelin injections.

Furthermore, for women over 50, ipamorelin is an excellent choice for improving their quality of deep sleep at night.

‘The ability of the growth hormone secretagogue (GHS) Ipamorelin to counteract the catabolic effects of glucocorticoid (GC) on skeletal muscles and bone was investigated in vivo in an adult rat model. 

Groups of 8-month-old female rats were injected subcutaneously for 3 months with GC (methylprednisolone) 9 mg/kg/day or GHS (Ipamorelin) 100 microg/kg three times daily, or both GC and GHS in combination. 

The maximum tetanic tension of the calf muscles was determined in vivo in a materials testing machine. 

The maximum tetanic tension was increased significantly, and the periosteal bone formation rate increased four-fold in animals injected with GC and GHS in combination, compared with the group injected with GC alone. 

In conclusion, the decrease in muscle strength and bone formation found in GC-injected rats was counteracted by simultaneous administration of the growth hormone secretagogue.”

CJC-1295

CJC-1295 is a synthetic long-acting growth hormone-releasing hormone (GHRH) analog that binds to specific receptors in the pituitary gland, stimulating the release of GH. 

Elevated GH levels lead to increased protein synthesis, more muscle mass, improved strength, and faster fat loss.

CJC-1295 also sustains GH secretion over extended periods, which supports ongoing fat loss and muscle growth even at rest.

When used together, CJC-1295 and ipamorelin offer the same benefits as either one in isolation… BUT the benefits are more pronounced than when either peptide is used in isolation.

This makes the CJC-1295 and Ipamorelin stack one of the most effective peptide combinations for boosting muscle growth and promoting fat loss.

A study published in the American Journal of Physiology-Endocrinology and Metabolism discovered that GHRHKO mice treated daily with CJC-1295 achieved normalized body weight and length after just 5 weeks:

“In children with isolated growth hormone (GH) deficiency, GH-releasing hormone (GHRH) can be effective for promoting growth. 

However, its short half-life limits its therapeutic use. 

To address this limitation, CJC-1295, a synthetic GHRH analog, was tested in mice lacking the GHRH gene. 

Three groups of 1-week-old GHRHKO mice were treated with CJC-1295 at intervals of 24, 48, and 72 hours for 5 weeks. 

Daily CJC-1295 treatment normalized body weight and length, while mice treated every 48 and 72 hours showed improved growth but not complete normalization. 

Bone length remained normal in animals treated every 24 and 48 hours. CJC-1295 also increased pituitary RNA and GH mRNA levels, indicating somatotroph cell proliferation. 

Overall, once-daily CJC-1295 administration effectively maintained normal body composition and growth in GHRHKO mice, with less frequent dosing being less effective.”

BPC-157

BPC-157, or “Body Protection Compound 157”, is a peptide made up of 15 amino acids that is isolated from human gastric juices. 

It’s known for its stability in the harsh acidic environment of the stomach. 

Your body produces small amounts of BPC-157 to initiate several beneficial processes in the digestive system. 

BPC-157 also stimulates the formation of new blood vessels via angiogenesis, which is crucial for healing and cell regeneration. 

This makes it particularly promising for speeding up the recovery of soft tissue injuries (muscle, tendon, ligament, etc.), helping in faster healing and recovery.

A study published in the Journal of Orthopaedic Research found that injecting BPC-157 led to faster muscle healing and complete restoration of function in rats with muscle crush injuries over a 72-day period:

“We report complete transection of major muscle and the systemic peptide treatment that induces healing of quadriceps muscle promptly and then maintains the healing with functional restoration. 

Initially, stable gastric pentadecapeptide BPC 157 (GEPPPGKPADDAGLV, M.W. 1419, PL-10, PLD-116, PL 14736 Pliva, Croatia; in trials for inflammatory bowel disease; wound treatment; no toxicity reported; effective alone without carrier) also superiorly accelerates the healing of transected Achilles tendon. 

Regularly, quadriceps muscle completely transected transversely 1.0 cm proximal to patella presents a definitive defect that cannot be compensated in rats. 

BPC 157 (10 microg, 10 ng, 10 pg/kg) is given intraperitoneally, once daily; the first application 30 min post transection, the final 24 h before sacrifice. 

It consistently improves muscle healing throughout the whole 72-day period. 

Improved are: (i) biomechanic (load of failure increased); (ii) function (walking recovery and extensor postural thrust/motor function index returned toward normal healthy values); (iii) microscopy/immunochemistry [i.e., mostly muscle fibers connect muscle segments; absent gap; significant desmin positivity for ongoing regeneration of muscle; larger myofibril diameters on both sides, distal and proximal (normal healthy rat-values reached)]; (iv) macroscopic presentation (stumps connected; subsequently, atrophy markedly attenuated; finally, presentation close to normal non injured muscle, no post surgery leg contracture). 

Thus, post transection healing-consistently improved-may suggest this peptide therapeutic application in muscle disorders.”

MK-677 (Ibutamoren)

MK-677, also known as Ibutamoren, is a growth hormone secretagogue that mimics ghrelin’s effects in the body by binding to growth hormone secretagogue receptors (GHSR) in the brain.

By stimulating the pituitary gland, MK-677 triggers a peak in GH release. 

This in turn elevates levels of GH and IGF-1 in the bloodstream, leading to the same outcomes seen with Ipamorelin and CJC-1295.

Specifically: Enhanced muscle growth, faster fat loss, and improved body composition.

A study published in The Journal of Clinical Endocrinology & Metabolism found that obese men who received oral MK-677 treatment for 8 weeks experienced an increase in fat-free mass and had an elevated basal metabolic rate (BMR).

(BMR = the rate at which the body expends energy while at rest)

“The study was double-blind, randomized, parallel, and placebo-controlled. 

It included 24 obese males between the ages of 18 and 50 with body mass index (BMI) greater than 30 kg/m2 and waist/hip ratios greater than 0.95. 

For eight weeks, the participants were given MK-677 25 mg or a placebo. 

Results showed that MK-677 treatment increased serum insulin-like growth factor I (IGF-I) levels by approximately 40% compared with placebo. 

Serum IGF-binding protein-3 levels were also significantly higher. 

The initial dose of MK-677 significantly increased GH and PRL (peak and area under the curve values). 

Significant increases in GH and PRL managed to remain after 2 and 8 weeks of treatment. 

On the other hand, the increases in GH and PRL after the initial dose were significantly greater than the increases seen after multiple doses. 

Cortisol levels in the blood and urine did not increase after 2 or 8 weeks. 

When measured with dual-energy X-ray absorptiometry, the subjects treated with MK-677 had a significant increase in fat-free mass. 

Active therapy had no discernible effect on total or visceral fat.

The basal metabolic rate also increased significantly after 2 weeks of MK-677 treatment but not after 8 weeks. 

Fasting glucose and insulin concentrations remained unchanged, but an oral glucose tolerance test demonstrated impaired glucose homeostasis (blood sugar balance) at 2 and 8 weeks. 

The study concluded that a two-month treatment with MK-677 in healthy obese males caused a sustained increase in serum levels of GH,IGF-I, and IGF-binding protein-3. 

The treatment also caused a sustained increase in fat-free mass and basal metabolic rate.”

IGF-1 LR3

IGF-1 LR3, or insulin-like growth factor-1 long R3, is a modified and more potent version of the regular IGF-1 that your body naturally makes.

It has a longer half-life than regular IGF-1, meaning it stays active in your system for a longer time. 

What makes IGF-1 LR3 unique is its ability to boost muscle growth and repair. 

It helps increase protein synthesis, speeding up muscle development while slowing the rate of muscle breakdown.

IGF-1 LR3 also helps your body retain nitrogen and protein, which are crucial for muscle development. 

Last but not least, IGF-1 LR3 also helps ramp up GH production.

Since it directly stimulates muscle growth, many bodybuilders inject it into the specific muscle groups they want to target for more localized gains.

Research published in the Journal of Trauma and Acute Care Surgery reported that combining exogenous GH with IGF-1 over an 8-week period significantly enhanced lean muscle growth, total body weight, and wound healing in burned rats

This combination was found to be more effective than administering GH or IGF-1 separately:

“Rats, each weighing 440-470 g, were given a 35% total body surface area, full-thickness scald burn and divided into four groups to receive placebo (burned controls), bovine GH (2.5 mg/kg/day), IGF-1 (2.0 mg/kg/day), or bovine GH plus rhIGF-1 (2.5 + 2.0 mg/kg/day), respectively, for 8 weeks.

As a result, total body weight gain after 8 weeks averaged 110 g for GH plus IGF-1 compared with gains of 49 and 11 g for GH or IGF-1 alone, respectively. 

Burned controls lost 24 g. 

Metabolic rates were significantly reduced in all groups receiving growth hormones. 

Gastrocnemius muscle dry weight was significantly increased in those receiving GH plus IGF-1 compared with GH and IGF-1 alone or burned controls (p < .01).

This data shows that GH plus IGF-1 synergistically increased lean muscle weight, total body weight, and was more effective in re-epithelialization of the burn wound than either GH or IGF-1 alone.”

MGF (Mechano Growth Factor)

MGF is a specialized form of IGF-1 that aids in muscle repair and recovery after exercise. 

It enhances muscle growth in targeted areas and reduces exercise-induced muscle damage, accelerating recovery so you can train more intensely and frequently. 

PEG-MGF, on the other hand, is a modified version of MGF, with polyethylene glycol (PEG) attached. 

This modification prolongs its effects from just a few minutes to as long as 48-72 hours. 

PEG-MGF does everything ordinary MGF does while also speeding up the body’s rate of natural healing, promoting nitrogen retention and upregulating protein synthesis. 

A study published in Biotechnology Letters on SpringerLink found that rats treated with MGF showed improved healing of injured Achilles tendons:

“A significantly increased Achilles functional index was found in the MGF-C25E-treated tendons relative to the saline-treated tendons. 

Macroscopic observation showed that the repair of MGF-C25E-treated (1000 ng/ml) tendons was superior to that of the saline-treated tendons. 

The mean value of the stiffness of the repaired tendons was significantly increased in the MGF-C25E-treated (1000 ng/ml) tendons relative to the saline-treated tendons. 

Moreover, histological analysis provided evidence of tissue remodeling and cell alignment.

This proves that MGF-C25E plays an important role in promoting healing of injured rat tendon and could be used as a potential therapeutic factor for tendon repair.”

GHRP-6 and GHRP-2

GHRP-2 and GHRP-6 are growth hormone-releasing peptides (GHRPs) that stimulate the hypothalamus and pituitary gland to release GH. 

By increasing GH secretion, these peptides boost protein synthesis, allowing the body to build and repair muscle tissue more efficiently. 

This results in faster muscle hypertrophy and improved overall muscle development.

In addition to promoting muscle growth, GHRPs help in fat loss by encouraging lipolysis (the breakdown of stored fat into fatty acids). 

This improves caloric burn during both rest and vigorous exercise, leading to a leaner and more defined physique.

Furthermore, GHRP-2 and GHRP-6 support post-exercise recovery by facilitating the repair and regeneration of damaged muscle fibers. 

Which allows you to maintain a more consistent and intensive training regimen since your body doesn’t require as much downtime to recover.

A study in PLOS ONE revealed that combining growth hormone-releasing peptide-2 (GHRP-2) with cysteamine (CS) significantly boosted growth in yaks with growth retardation, leading to notable improvements in muscle development and hormone levels:

“The objective of this study was to investigate the effects of growth hormone-releasing peptide-2 (GHRP-2) and cysteamine (CS) administration on growth performance in yaks with growth retardation and try to elucidate its regulatory mechanisms. 

Trial 1, thirty-six 1-year-old Qinghai high plateau yaks (body weight 38–83.2 kg) were randomly chosen for body weight and jugular blood samples collection. 

The relationship between body weight and serum GHRH (P < 0.05, R = 0.45), GH (P < 0.05, R = 0.47), IGF-1 (P < 0.05, R = 0.62) was significantly correlated in yaks colonies with lighter body weights. 

Trial 2, fifteen 1-year-old Qinghai high plateau yaks with growth retardation (average body weight 54.8 ± 8.24 kg) were randomly selected and assigned to negative control group (NG), GHRP-2 injection group (GG) and cysteamine feeding group (CG), with 5 yaks per group. 

Another five 1-year-old Qinghai high plateau yaks with normal growth performance (average body weight 75.3 ± 2.43 kg) were selected as positive control group (PG). 

The average daily gain (ADG) of the GG and CG were significantly higher than those in the PG and NG (P < 0.05). Both GHRP-2 and CS administration significantly enhanced the myofiber diameter and area of skeletal muscle (P<0.05). 

GHRP-2 significantly enhanced the serum GH and IGF-1 levels (P < 0.05), and up-regulated GHR, IGF-1 and IGF-1R mRNA expression in the liver and skeletal muscle (P < 0.05), enhanced the mRNA expression of PI3K, AKt and mTOR in the skeletal muscle (P<0.05). 

CS significantly reduced the serum SS levels and the hypothalamus SS mRNA expression (P < 0.05), and enhanced GHR and IGF-1 mRNA expression in the liver (P < 0.05), decreased the mRNA expression of muscle atrophy F-box (Atrogin-1) and muscle ring finger 1 (MuRF1) mRNA (P < 0.05). 

Therefore, growth retardation in yaks was primarily due to somatotropic axis hormones secretion deficiency. 

Both GHRP-2 and CS administration can accelerate growth performance and GH, IGF-1 secretion in yaks with growth retardation. 

GHRP-2 enhanced muscle protein deposition mainly by up-regulating the protein synthesis pathways, whereas CS worked mainly by down-regulating the ubiquitin-proteasome pathway.”

Another study published in BMB Reports found that the new GHRP-6-biotin conjugate improved muscle function in cultured cells by boosting key muscle proteins, IGF-1, and collagen type I.

This was in addition increasing energy levels and enzyme activity, indicating that it may help enhance muscle health:

“Based on the potential beneficial effects of growth hormone releasing peptide (GHRP)-6 on muscle functions, a newly synthesized GHRP-6-biotin conjugate was tested on cultured myoblast cells. 

Increased expression of myogenic marker proteins was observed in GHRP-6-biotin conjugate-treated cells. 

Additionally, increased expression levels of insulin-like growth factor-1 and collagen type I were observed. 

Furthermore, GHRP-6-biotin conjugate-treated cells showed increased metabolic activity, as indicated by increased concentrations of energy metabolites, such as ATP and lactate, and increased enzymatic activity of lactate dehydrogenase and creatine kinase. 

Finally, binding protein analysis suggested few candidate proteins, including desmin, actin, and zinc finger protein 691 as potential targets for GHRP6-biotin conjugate action. 

These results suggest that the newly synthesized GHRP-6-biotin conjugate has myogenic stimulating activity through, at least in part, by stimulating collagen type I synthesis and several key proteins. 

Practical applications of the GHRP-6-biotin conjugate could include improving muscle condition.”

It is ALWAYS smart to with the lower dose to gauge how your body responds and then gradually adjust the dose and/or frequency as needed.

Additionally, remember peptides are best used in a cyclical fashion; a common approach is an 8 to 12-week cycle followed by a break for an equal amount of time. 

This will allow you to avoid the dreaded peptide antibody buildup and ensure these peptides remain effective year-round.

As for the best method of administration, subcutaneous/injection will always be the way to go 99% of the time.

If you choose this method, use the correct syringes, follow the proper steps to prevent infection, and rotate injection sites to avoid irritation. 

(NOTE: Some peptides can be taken orally or applied as topical creams, but these are rare exceptions and not the rule)

Before starting any peptide regimen, consult with your doctor for personalized advice and closely monitor your body’s response to make adjustments as needed based on your progress and any side effects.

As I have already stated in numerous recent articles, NONE of the above bodybuilding peptides are practically relevant in the context of reliably inducing muscular hypertrophy.

Many bodybuilders will use these peptides and over-state the role they played in enhancing their physique, while simultaneously downplaying other factors (diet, training, sleep, other anabolic agents, etc.).

This is why so many people are hopelessly confused about what works and what doesn’t.

Yes, I AM aware of the many animal studies and cell studies showing these peptides may be effective for building skeletal muscle mass.
