Every time someone calls raw follistatin a peptide you can buy and inject, I know exactly what level of education I’m dealing with.
Native follistatin is a 344-amino-acid glycoprotein with complex folding properties, glycosylation sites, and isoform-specific binding activity.
A long-winded way of saying it is NOT a peptide!
To elucidate this difference further:
- A glycoprotein consists of sugar groups attached to a protein or large polypeptide (>50 amino acids, usually)
- A peptide consists of 2-50 amino acids connected via peptide binds
This is not a semantic argument: While this seems like a small difference, it’s more than enough to dictate how the molecule folds in three-dimensional space and how it behaves in your body.
Coming back to the conversation at hand the most studied isoforms are FS-288 and FS-315… both of which behave very differently depending on tissue context and binding partners.
Here is where things get interesting, as these details underpin the engineering problem FLGR-242 was built to solve.
FLGR-242 is a fragmented and modified version of Follistatin-344 fused to a proprietary albumin-binding construct, which makes it a true engineered peptide with a molecular weight around 40 kDa.
So when you understand that native follistatin is a glycoprotein, you also understand why crude “follistatin” off a random vendor was – and is – never going to work.
Myth #2: Follistatin Just Builds Muscle
Yes, follistatin inhibits myostatin, the transforming growth factor-beta (TGF-β) superfamily member that acts as the molecular brake on skeletal muscle growth.
When myostatin is neutralized, the SMAD2/SMAD3 cascade goes quiet, satellite cells activate more readily, and muscle hypertrophy follows, a mechanism documented in animal models that goes back to McPherron and Lee’s 1997 work published in Nature.
But native follistatin also binds to — and antagonizes — several other signaling molecules.
It binds Activin A and Activin B (regulation of energy metabolism and reproductive function)
It also binds bone morphogenic proteins BMP-2, BMP-4, and BMP-7 (regulation of bone formation, adipogenesis, and cellular differentiation)
And it binds to growth differentiation factor 11 (GDF-11), which is involved in aging biology and organ regeneration.
Native follistatin is most accurately thought of as a systemic signaling regulator, and its broad binding profile is exactly why every earlier attempt to inhibit myostatin created off-target effects.
FLGR-242 sidesteps this by starting from the FS I-I isoform, which retains full myostatin-neutralizing power while dramatically reducing activin binding.
Translation: You get the muscle-building signal without the hormonal disruption.
Myth #3: Follistatin Supplements Work
Walk into any supplement store, online or brick-and-mortar, and you will find capsules and powders claiming to deliver follistatin-like results.
Since follistatin is a large glycoprotein, and oral ingestion subjects it to proteolytic degradation in the GI tract before any bioactive form reaches systemic circulation… it just simply doesn’t work as a pill you can pop and swallow.
Furthermore, there is no credible evidence that orally consumed follistatin crosses the gut barrier intact and exerts meaningful systemic effects.
What these products typically contain are epicatechins, i.e. compounds that MAY modestly nudge myostatin levels via upstream pathways.
Epicatechin has its own limited data, but it is NOWHERE NEAR follistatin in terms of outcomes produced, so labeling it as such is misleading at best and predatory at worst.

Myth #6: Higher Follistatin Equals Unlimited Muscle Growth

- Androgen receptor density

- mTORC1 signaling
