Following down-regulation with a gonadotrophin-releasing hormone agonist (GnRHa), patients were randomized to receive GRF (500 microg twice daily; n = 96) or placebo (n = 100) in addition to follicle stimulating hormone (FSH); treatment was continued until human chorionic gonadotrophin was given, or for a maximum of 14 days. 

GRF had no significant effect on the mean number of follicles with a diameter of >/=16 mm (GRF: 3.26 +/- 2.29; placebo: 3.27 +/- 2.30; P = 0.95), the number of FSH ampoules required to achieve ovarian stimulation (GRF: 55.2 +/- 16. 4; placebo: 54.9 +/- 17.2; P = 0.50), or on secondary measures of ovarian response and treatment outcome. 

There were, however, significant increases in circulating growth hormone (GH) and insulin-like growth factor (IGF)-1 concentrations. 

GRF was well tolerated. It is concluded that, despite producing significant increases in GH and IGF-1, concomitant treatment with GRF does not improve the ovarian response to FSH in poorly responsive women undergoing IVF.”

Subjects of both groups were not different at baseline. 

After 6 months of therapy, muscle strength in the bench press responsive muscles did not increase in both groups and showed a statistically significant increase in the leg press responsive muscles in the GH group. 

Our study demonstrated an increase in muscle strength in the lower body part after GH therapy in healthy men. 

This finding must be considered and tested in frail older populations, whose physical incapacity is primarily caused by proximal muscle weakness.”

The mechanical test showed that strain at maximum load, maximum load, and relative failure energy increased with increasing doses of biosynthetic hGH. In the group receiving 8.33 mg.kg-1.day-1, skin collagen content per surface area and skin collagen concentration in percent of dry weight was increased, whereas the fat concentration in percent of dry weight was decreased. 

Also when correcting the mechanical data for cross-sectional area, a positive correlation between dose and relative failure energy was found. 

When dividing the mechanical data by collagen content per surface area of the skin, the maximum stress and relative failure energy reached the highest value at a dose of 1.10 mg.kg-1.day-1. 

The study shows that biosynthetic hGH can induce an increase in collagen content and mechanical strength of intact rat skin which is dependent on the dose of biosynthetic hGH, and that the increase in mechanical strength cannot be explained only by an increased collagen content.”

“Thirteen-week-old female Sprague-Dawley rats were given IPA (0.5 mg/kg per day; n=7), GHRP-6 (0.5 mg/kg per day; n=8), GH (3.5 mg/kg per day; n=7), or vehicle administered continuously s.c. via osmotic minipumps for 12 weeks. 

The animals were followed in vivo by dual X-ray absorptiometry (DXA) measurements every 4th week. 

After the animals were killed, femurs were analysed in vitro by mid-diaphyseal peripheral quantitative computed tomography (pQCT) scans. 

After this, excised femurs and vertebrae L6 were analysed by the use of Archimedes’ principle and by determinations of ash weights. 

All treatments increased body weight and total tibial and vertebral BMC measured by DXA in vivo compared with vehicle-treated controls. 

However, total BMC corrected for the increase in body weight (total BMC:body weight ratio) was unaffected. 

Tibial area bone mineral density (BMD, BMC/area) was increased, but total and vertebral area BMDs were unchanged. 

The pQCT measurements in vitro revealed that the increase in the cortical BMC was due to an increased cross-sectional bone area, whereas the cortical volumetric BMD was unchanged. 

Femur and vertebra L6 volumes were increased but no effect was seen on the volumetric BMDs as measured by Archimedes’ principle. 

Ash weight was increased by all treatments, but the mineral concentration was unchanged. We conclude that treatment of adult female rats with the GHSs Ipamorelin and GHRP-6 increases BMC as measured by DXA in vivo. 

The results of in vitro measurements using pQCT and Archimedes’ principle, in addition to ash weight determinations, show that the increases in cortical and total BMC were due to an increased growth of the bones with increased bone dimensions, whereas the volumetric BMD was unchanged.”

Ipamorelin can also be used as a peptide for injury recovery because it helps repair damaged tissue caused by physical trauma

This happens via boosting collagen synthesis and providing structural/biochemical support in the cells surrounding the injured area:

“Members of the transforming growth factor beta (TGFβ) superfamily of secreted factors play essential roles in nearly every aspect of cartilage formation and maintenance. 

However, the mechanisms by which TGFβs transduce their effects in cartilage in vivo remain poorly understood. 

Mutations in several TGFβ family members, their receptors, extracellular modulators, and intracellular transducers have been described, and these usually impact the development of the cartilaginous skeleton. 

Furthermore, genome-wide association studies have linked components of the (TGFβ) superfamily to susceptibility to osteoarthritis. 

This review focuses on recent discoveries from genetic studies in the mouse regarding the regulation of TGFβ signaling in developing growth plate and articular cartilage, as well as the different modes of crosstalk between canonical and noncanonical TGFβ signaling. 

These new insights into TGFβ signaling in cartilage may open new prospects for therapies that maintain healthy articular cartilage.”

Improved Sleep Quality, Energy Levels, and Mood

“Fourteen patients with untreated GHD of confirmed or likely pituitary origin, aged 22–74 years, participated in the study. 

Patients with associated hormonal deficiencies were on appropriate replacement therapy. 

Polygraphic sleep recordings, with bedtimes individually tailored to habitual sleep times, were performed after 4 months on rhGH or placebo.

Valid data were obtained in 13 patients. At the end of the rhGH treatment period, patients had a shorter sleep period time than at the end of the placebo period (479±11 vs 431±19 min respectively; P=0.005), primarily due to an earlier wake-up time, and a decrease in the intensity of SWS (delta activity) (559±125 vs 794±219 μV2 respectively; P=0.048).

Four months of rhGH replacement therapy partly reversed sleep disturbances previously observed in untreated patients. 

The decrease in delta activity associated with rhGH treatment adds further evidence to the hypothesis that the excess of high-intensity SWS observed in untreated pituitary GHD patients is likely to result from overactivity of the hypothalamic GHRH system due to the lack of negative feedback inhibition by GH.”

Similarly, rats given the growth hormone-releasing peptide had activated sleep regulatory neurons in the brain, resulting in better sleep:

“Intracerebroventricular administration of GHRH significantly increased (P < 0.001) the amount of NREM sleep compared with the control group of rats at the beginning of the active/dark period. 

A significant increase (P < 0.001) in the EEG SWA was also observed in the GHRH intracerebroventricularly injected rats compared with the control group. 

However, REM sleep amount did not change significantly (P = 0.311) following intracerebroventricular injection of GHRH. 

The latency of NREM sleep onset decreased significantly (P < 0.05) after GHRH treatment (22.50 ± 2.68 min) compared with control saline (34.17 ± 4.57 min) treatment group during the dark period.”

While Ipamorelin is generally considered safe, it has yet to receive approval from the FDA or international regulatory bodies for human use
